Abstract

Summary: The ultrastructure of liver tissues from guinea pigs, rabbits, mice, and rats was examined after chronic administration of the anorectic drug chlorphentermine. While hepatocytes of rats remained largely unaltered, those of guinea pigs, rabbits and mice contained numerous membrane-limited inclusion bodies with a lamellar, a reticular or a crystallinelike internal structure. Similar inclusions were found in increased numbers within Kupffer cells of rats, guinea pigs and mice. The cytological alterations are interpreted to result from an abnormal intralysosomal accumulation of phospholipids or other lipids. The findings are taken as supporting the concept of a drug-induced lipidosis.
